Application of Uricase Isolated from Bacillus subtilis SP6 in Uric Acid Assay Diagnostic Kit

Uricase enzyme is a major component of the diagnostic kit for estimation uric acid diagnosis, monitoring and treatment gout joint inflammations. Uric Acid/Uricase assay simple measuring concentrations in biological samples such as serum, plasma, urine without any need pretreatment. The level uricase activity was detected crude extract some animal liver tissues, plant leaves microbial source (Bacillus subtilis SP6 bacteria) expressed specific (unit/mg protein). It found difficult to produce high yield short time from or tissues stability both sources still unclear; therefore, it isolated Bacillus SP6. extraction procedure involved, isolation bacterial cells, determining intracellular extracellular fractions, pooling fractions ammonium sulfate precipitation which seemed be convenient since 74.7 % recovered. applied preparation that sensitive comparable with commercially available ones.
